A study by Galea et al. in the Journal of Cell Biology identifies a role for the Golgi in regulating DNA repair protein availability. Screening and siRNA validation confirmed 163 proteins with dual Golgi-nuclear localization, including a network of DNA damage response factors.
Depletion of the Golgi scaffold giantin caused premature nuclear accumulation of RAD51C, reduced homologous recombination efficiency, and increased genomic instability. These findings support a model in which Golgi-associated retention helps coordinate repair factor availability.
The researchers used Invent Biotechnologies’ Minute™ Golgi Apparatus Enrichment Kit (GO-037) for Golgi enrichment, assessing fractions with GM130 and TGN46. This preparation step formed part of a broader workflow combining localization analysis, biochemical fractionation, and functional repair assays.
GO-037 uses spin-column technology and preferential precipitation to recover Golgi-enriched material and a separate Golgi secretory vesicle fraction without ultracentrifugation. For researchers investigating Golgi-associated signaling or protein redistribution, it provides a practical route to material for downstream biochemical analysis.
